چکیده

Blocks of the kidney and other organs of the guinea pig fixed with glutaraldehyde and treated with tannic acid and osmium tetroxide could, without any coating, be observed in the scanning electron microscope. Undesirable charging effect was inconspicuous when accelerating voltage 5-25kV was used. A scanning image of the renal glomerulus was shown and the details of the method were described. The merit of this method is avoidance of the metal coating which obscures the details of the specimen surface.

Direct Observation of Unstained Biological Specimens in Water by the Frequency Transmission Electric-Field Method Using SEM

Scanning electron microscopy (SEM) is a powerful tool for the direct visualization of biological specimens at nanometre-scale resolution. However, images of unstained specimens in water using an atmospheric holder exhibit very poor contrast and heavy radiation damage. Here, we present a new form of microscopy, the frequency transmission electric-field (FTE) method using SEM, that offers low rad...

High-Contrast Observation of Unstained Proteins and Viruses by Scanning Electron Microscopy

Scanning electron microscopy (SEM) is an important tool for the nanometre-scale analysis of the various samples. Imaging of biological specimens can be difficult for two reasons: (1) Samples must often be left unstained to observe detail of the biological structures; however, lack of staining significantly decreases image contrast. (2) Samples are prone to serious radiation damage from electron...

The selective catalytic reduction of NO over Ce0.3TiOx-supported metal oxide catalysts.

A Ce0.3TiOx oxide carrier was synthesized via a sol-gel process, and Ce0.3TiOx supported metal (M=Cd, Mn, Fe, W, Mo) oxide catalysts were prepared by the method of incipient-wetness impregnation. The catalysts were characterized by means of X-ray diffraction (XRD), Brunauer-Emmett-Teller (BET) analysis, scanning electron microscopy (SEM), transmission electron microscopy (TEM), Fourier transfor...

Immunogold labelling for scanning electron microscopy.

Scanning electron microscopes are useful biological tools that can be used to image the surface of whole organisms, tissues, cells, cellular components and macromolecules. Processes and structures that exist at surfaces can be imaged in pseudo or real 3D at magnifications of anything from about x10 to x1,000,000. Therefore a whole multicellular organism, such as a fly, or a single protein embed...

Response Behaviour of a Hydrogen Sensor Based on Ionic Conducting Polymer-metal Interfaces Prepared by the Chemical Reduction Method

A solid-state amperometric hydrogen sensor based on a protonated Nafion membrane and catalytic active electrode operating at room temperature was fabricated and tested. Ionic conducting polymer-metal electrode interfaces were prepared chemically by using the impregnation-reduction method. The polymer membrane was impregnated with tetra-ammine platinum chloride hydrate and the metal ions were su...
